It functions much like 'even though,' 'despite,' or 'and yet' in English, highlighting a contradiction between two facts.
Since くせ is treated as a noun, くせに follows the attributive form of the word that precedes it.

- あんなに体が大きいくせに、力は弱いんだ。Despite being so big, he's surprisingly weak.
