It functions much like 'even though,' 'despite,' or 'and yet' in English, highlighting a contradiction between two facts.
Since くせ is treated as a noun, くせに follows the attributive form of the word that precedes it.
- 「手伝う」って言ったくせに、何もしてくれないじゃん。You said you'd help, and yet you haven't done a single thing.
- あんなに体が大きいくせに、力は弱いんだ。Despite being so big, he's surprisingly weak.

The word 癖 has evolved to carry a negative nuance. Initially, it simply referred to a natural inclination or a preference for something. By attaching the case marking particle に, the grammar pointed out a contradiction: when state (A) is the expected or proper state, the failure of (B) to align with it is seen as undesirable or inappropriate.
